Kay, 27, opted out of the third year of his Perth Wildcats contract in May to assess his opportunities outside of the NBL (he had a European get-out clause in his contract).
The Boomer's desire to make his Olympic debut, in 2021, was the main reason he left the NBL for Spain's Liga ACB.
In a video posted on Real Betis' Twitter account, Kay said he was "very excited" about the upcoming season.
"[I] can't wait to get over there, get started and win some games. See you all soon," he said.
In May, Kay told the Leader that "it's always been a goal of mine to push myself and try and get into a European team.
"I've been looking to do this for a while now. And this just gives me the opportunity to see what's out there, and hopefully things fall into place and a good opportunity presents itself."
Kay's two seasons at the Wildcats earned him back-to-back championships and All-Star First Team honours both seasons.
He represented Australia at the 2019 FIBA World Cup in China.
In 2020, he was named the Wildcats' players' player and defensive player.
Perth Wildcats chief executive Troy Georgiu said on Wildcats.com.au that he was pleased for Kay.
"It's rewarding to see someone who's been an integral part of our program be afforded an opportunity to achieve a personal goal," he said.
"I'm looking forward to watching Nick continue to grow in Europe, and am excited to hopefully see him in the green and gold at the Tokyo [Olympic] Games."
