Mel Behrens-Macaulay and Andrew Macaulay are riding from Hervey Bay to Sydney, via the New England Highway, in The Bloody Long Ride.
Ms Behrens-Macaulay has the disease, which has rendered her blind in one eye, caused severe muscle fatigue and heart problems.
“The Bloody Long Ride is a celebration of life and enjoying what I can do today, irrespective of what health challenges are ahead,” she said.
Australian Mitochondrial Disease Foundation CEO Sean Murray said more than 120,000 Australians are at risk of developing mitochondrial disease.
“At least one Australian child born each week will develop a severe form of mitochondrial disease, and half will die in childhood.”
The pair will also join 2,000 people in Sydney for The Bloody Long Walk, on November 13.
