NSW Health confirmed the latest sample taken on Tuesday found fragments of the COVID-19 virus, and Tamworth Regional Council expected a follow up test would provide new results on Saturday.
"We will have another sewer test ... sent to Sydney [on Thursday afternoon] and the results of that will be available on Saturday," mayor Col Murray said.
No new cases of COVID-19 were recorded in the region in the 24 hours to 8pm on Thursday, meaning the city has remained free of the virus since April last year.

Authorities remain concerned about the COVID-19 detection in the sewage considering there are no known cases in the area, and the plant services about 45,000 people locally.
The detection comes three weeks after a positive COVID-19 case unknowingly visited Tamworth, though no cases in the area have been confirmed since.
Tamworth MP Kevin Anderson said it was a "wake up call" for people to get tested and vaccinated.
Testing levels across the city have taken a dive and data from Hunter New England Health (HNEH) shows in the past few days in Tamworth, 387 people were tested on August 20; 216 were swabbed the next day; and 211 were swabbed on August 22.
A COVID-19 swab can be sought at the Plain Street drive-through, the Tamworth hospital, the respiratory clinic and Douglass Hanly Moir pathology.
It's understood the AELEC drive-through may reopen if demand requires.
The state recorded 882 cases of community transmission on Friday, and two of those cases were in the HNEH district.
