Police said the medium rigid truck with a back trailer was travelling east when it appears to have rolled at about 2.30pm on Thursday.
The incident happened about 30km south-east of Bingara at Back Creek.
A 56-year-old male driver and 23-year-old male passenger, both from Victoria, suffered minor injuries and were treated by paramedics.
Police said the truck was carrying a range of chemicals used for rural purposes.
The chemical spill has forced the closure of the road since the time of the accident.
Fire and Rescue NSW firefighters including the Hazmat team are still on the scene assisting with the clean-up.
The road will be reassessed later on Friday with police anticipating that it may be re-opened by the late afternoon.
Updates on the road closure are available on the Live Traffic website.
