Little Kindy Childcare already operates a 41-child centre in East Tamworth Medical Centre.
It's one of five centres, in Newcastle, Broadmeadow, Muswellbrook and South Australia.
Plans for the new South Tamworth centre published by Tamworth Regional Council last week show the centre would occupy a currently-vacant lot at 1 Edward Street.
Tamworth-based Director, Arzal Arzal, said the Tamworth site was proving too popular to keep up with demand, forcing expansion.

Several of their existing clients live on the south side of town, and an extra site would make things easier for them, he said.
He said the city is also just a great place to invest.
"I'm in love with Tamworth," he said.
"I believe in Tamworth's potential growth. I bring my family here and we love it.
"From a business perspective, we can see a very good response from the parents, from the family, from the community.
"We are confident that Tamworth will be a great community to support our expansion,"
The centre would cater for as many as 38 children and employ between seven and eight full time staff, plus casuals, he said.
If approved construction will cost an estimated $719,237.61.
It would take about 9 to 12 months to build, and could be open by 2023, assuming no planning hurdles.
Mr Arzal said there were many benefits to smaller childcare centres
"It's kind of like a boutique ... centre," he said.
"It's going to be 38 children. We love small centres so we can focus more on the family. It's kind of like a family-oriented centre."
Little Kindy isn't the only daycare provider to take note of the local shortage.
In August, the Ekidna early learning chain revealed plans to convert the old St Nicholas Catholic school campus into an 85-child early learning centre.
