In a sprint finish, the division one Rauland rider held off Edwin Britts of the Dulwich Hill Bicycle Club - with both riders clocking 2hr 50min 38sec for the 100km journey.
Sean Whitfield, of the Canberra Cycling Club, finished 1.55sec adrift in third place. Like Britts, Whitfield competes for Oliver's Real Food Racing.
Whitfield also clocked the fastest time in finishing ninth in the Keegan Downes Memorial Sundowner Handicap Cycling Classic on Saturday.
Spenceley, of Lidcombe Auburn Cycle Club, had a previous best finish of fourth in the annual Gunnedah to Tamworth.
Winning the race was "awesome", he said, adding: "This race suits me."
"I've just been chipping away at the NRS [National Road Series] races and the state races," he also said.
"The dream always is to race over in Europe."
This year Spenceley won under-23 criterium silver at state championships.
On Sunday, he said he "outfoxed" Britts and Whitfield.
"I was pretty confident," he added.
"I was just biding me time, waiting for the right time to attack Edwin, because we dropped Sean with about 10km to go."
Armidale's Michael Harris finished in seventh place, 7min 5ec behind Spenceley, to lead the way for the New England contingent.
Luke Deasey was the best-placed Tamworth competitor (20th).
Another Tamworthian, Brendan Langenbaker, finished third in division five, while Gunnedah's William Martin and Tamworth's Isla Maidment came first and third in division six.
