With almost 50 vehicles registered already, the outback adventure is set to be one of the biggest ever.
It's the first time the event will start away from Tamworth with a farewell dinner in Broken Hill, event organiser Jeff Galbraith said.
"One of the great things about the Drover's Run is that it draws entrants from well outside our region," he said.
"This year we have four vehicles entered from Sydney and a number from Queensland which brings in much-needed fundraising from areas away from the New England North West."

The 15-day outback adventure takes drivers through some of Australia's most stunning country from August 4.
The almost 3700km journey travels through the Flinders Ranges, across the Simpson Desert to Big Red and into Birdsville before it returns home.
